    Protein A/G magnetic beads contain a recombinant Protein A/G that combines the IgG binding domains of both Protein A and Protein G. Protein A/G allows for the capture of immunoglobulins from a broader range of species and antibody isotypes than either Protein A or Protein G alone.

    The protein A/G magnetic beads provided by Alfa Chemistry have a large specific surface area, more binding sites, higher antibody binding capacity, and extremely low non-specific binding rates. They are typically used for isolating antibodies from serum, cell culture supernatants, or ascites, as well as for immunoprecipitation and co-immunoprecipitation of antigens from cell or tissue extracts.

    Recommended Buffers

    The following are only recommended buffer components. You can adjust the buffer formulation as needed. Please note that all reagents need to be prepared using DEPC-treated purified water.

    • Binding buffer: 10×PBS, 1% (v/v) TritonX-100, 0.01% (v/v) NP-40, 5% (v/v) Glycerol
    • Washing buffer: 50mM Tris-HCl, 0.1% (v/v) Tween-20, pH7.5
    • Elution buffer: 0.1M Glycine, 0.1% (v/v) Tween-20, pH2.5

    You Need to Know

    • Freezing of magnetic beads or any other inappropriate operation is prohibited.
    • Before using our products, make sure to thoroughly oscillate to maintain a uniform suspension of the beads.
    • To ensure optimal experimental results, please select antibodies with high specificity for the immunoprecipitation reaction.
    • Magnetic beads should be stored in the storage solution to prevent drying. Drying can cause loss of binding capacity and result in reduced performance.
    • Our products need to be used with magnetic separation equipment.
    • Our products are for research use only.

    Troubleshooting

    ProblemPossible CauseSolution
    The binding efficiency of the antibody to the magnetic beads is low.The binding efficiency of the magnetic beads with antibody is related to the species origin and subtype of the antibody.Check the affinity of antibody and the Protein A/G matrix (see appendix).
    The antibody subtype has a lower affinity for Protein A/G matrix.Increase the incubation time of the antibody with the magnetic beads (30-120 min), increase the pH of the binding buffer (8-9) and decrease the ionic strength (25-100 mM NaCl).
    Aggregation of magnetic beads occurs during storage or use.Improper storage of magnetic beads.Handle the magnetic beads according to the instructions in the manual.
    Freeze or centrifuge magnetic beads.
    Clumping of magnetic beads occurs during use.Prolonged placement of the magnetic beads in the magnetic field causes them to firmly bind together.Ultrasonic water bath treatment for 2 minutes can redisperse the magnetic beads. However, note that ultrasonic treatment may also cause the antibodies captured by the magnetic beads in the sample solution to detach, so this method should not be used after adding the sample and before elution.
